PT-141 is a non-selective agonist of melanocortin receptors, primarily melanocortin receptors 3 and 4 (MC3 & MC4), with the exception of melanocortin receptor 2 (MC2 – the ACTH receptor). Its bioavailability after subcutaneous injection is approximately 100%, maximal levels occur after approximately 1 hour, with a range of 0.5 to 1.0 hours, and plasma protein binding of PT-141 is approximately 21%.

Erectile function is the result of a coordinated interaction of the vascular, endocrine, and nervous systems, which results in cGMP-mediated increased blood flow to the penis, a concomitant decrease in venous outflow, and the production of an erection. ED caused by untreated endocrine disease, anatomic deformations of the penis, prostatic disorders, or evidence of clinically significant hepatic, renal, cardiovascular, psychiatric, or CNS disease, including stroke or spinal cord injury, were all exclusion criteria.
Prior to the enrollment of research subjects, a human ethics committee approved the study protocol and informed consent document for each study, and all volunteers provided written informed consent prior to their participation in the study.
